Understanding the effects of column temperature on high resolution glycan separations

A solid core 2.6 μm column affords lower backpressures than a fully porous sub-2 μm column. This lower backpressure allows users to experiment with column temperature to understand its impact on chromatographic separation.
In this study, it was found that lower column temperatures showed higher resolution chromatograms, with more glycan variant forms identified versus higher temperature experiments on four therapeutic monoclonal antibodies. The Accucore 150-Amide-HILIC column has demonstrated outstanding separation compared to a fully porous 1.7 μm competitor HILIC column.
