The colony formation assay is an in vitro cellular assay based on the ability of a single cell to proliferate into a colony, which is a hallmark of cancer cells. This assay is crucial to evaluate the potency of anti-cancer drugs and their clinical potential. Traditionally, colonies cultured in multi-well plates are fixed, stained with Crystal Violet colorimetric dye, then manually scored by eye. Below, we demonstrate an automated high-throughput method using Crystal Violet fluorescence to image, identify and quantify colonies.
